FlexiBac PTM

In insect cells, N-glycosylations are truncated due to the absence of several glycosyltransferases and synthases.

Our FlexiBac technology has been adapted to optimize Post-Translational Modifications (PTMs) such as N-glycosylation or proteolytic maturation. This new technology, FlexiBac PTM, enables the production of glycoproteins displaying different N-glycan structures, ranging from paucimannose to complex types (with terminal galactose or sialic acids).

Conversion from insect N-glycans to human complex N-glycans

The FlexiBac PTM technology involves the permanent integration of genes encoding glycosyltransferases and synthases into the viral DNA to complement the host cell’s enzymatic machinery.

Modified viral DNA to express the missing glycosyltransferases and synthases in insect cells

Characterization by Western Blot or Lectin Blot of a lysosomal protein produced (1) with our standard FlexiBac technology (mannosylated protein), (2) with our FlexiBac MPT technology (sialylated protein) or (3) in CHO cells (sialylated protein)

It is also possible to play on the proteolytic maturation of proteins by using, for example, a baculovirus which overexpresses a cellular furin.
